| Message | Product Description REAGEN™ Botulinum Toxin A Strip Test is a qualitative and rapid lateral flow assay designed to detect Botulinum Toxin A residues in meat, feed, milk, honey, milk powder, soil, vomit, feces, serum and water sample. This test is designed for field use or reference lab settings. The unique features of the kit are: Rapid strip test method – 3 minutes High sensitivity (80ppb) High reproducibility
Procedure Overview The assay uses a double antibodies sandwich colloidal gold based format. The sample is added to the test well to allow the Botulinum Toxin A binding protein on the NC membrane to engage with any Botulinum Toxin A present in the sample. The gold particles conjugated anti-Botulinum Toxin A will bind to the drug that are bind by the Botulinum Toxin A binding protein at the Test line (T-line), forming a signal (red line) at that position. The test is negative if no colored line appears in the T area.
Kit Contents, Storage and Shelf Life The REAGEN™ Botulinum Toxin A Strip test has the capacity for 48 deter-minations. The shelf life is 24 months when the kit is properly stored. Kit Contents Amount Storage Reaction strip 48 strips 4°C-25°C Manual 1 RT
Sample preparation
Meat/Feed/Sauce/Milk powder 1. Grind and mix a representative sample(according to accepted sample techniques) 2. Weight out 2 g of the ground sample and place into a suitable container. 3. Add 5ml stroke-physiological saline solution and shake for 10 minutes with a vortex. 4. Centrifuge sample for 5 minutes at 4000 rpm. 5. Use the 200μL supernatant per well for the test.
Honey/Milk/Serum 1. Take 0.1ml sample into a 2ml tube , add 0.4ml stroke-physiological saline solution and shake for 1 minutes with a vortex. 2. Use the 200μL supernatant per well for the test.
Soil/Vomit/Feces 1. Weight out 1 g of the ground sample and place into a suitable container. 2. Add 5ml stroke-physiological saline solution and shake for 10 minutes with a vortex. 3. Centrifuge sample for 5 minutes at 4000 rpm. 4. Use the 200μL supernatant per well for the test.
Water 1. Use the 200μL sample per well for the test.
Testing Protocol
1. Using scissors, carefully cut and remove the number of strips appropriate for the number of samples that will be tested and allow reagents to reach ambient temperature (25˚C +/- 5). Ensure that unused strips remain firmly capped. Promptly re-store the remaining components at 4˚C-25˚C. 2. Take 3 points( about 200ul ) of sample to the test well, time immediately. 3. After 3 min, visually examine the intensity of the signal at the T-line and C-line. Interpretation of results. Colored lines which appear after 15 minutes are not diagnostic and should be ignored |
